A Brief Overview
The Weber Lab at The Children's Hospital of Philadelphia and The University of Pennsylvania Perelman School of Medicine seeks an experienced research assistant to help develop approaches to enhance chimeric antigen receptor (CAR-T) cell therapeutics.
CAR-T cell therapies have revolutionized the cancer immunotherapy landscape by demonstrating remarkable clinical responses in patients with B cell malignancies. However, T cell exhaustion and poor persistence limit efficacy in patients and are major barriers to progress for the treatment of solid tumors. The Weber Lab is developing a suite of approaches (ex. overexpression of engineered proteins, CRISPR-mediated genetic deletion, and small molecule treatment) to transcriptionally and epigenetically reprogram T cells to enhance anti-tumor potency, while simultaneously uncovering molecular mechanisms that govern human T cell fate and function.
The candidate will design and perform pre-clinical studies that includes maintenance of human T cell and tumor cell cultures, T cell engineering (ex. viral production and T cell transduction), cloning and molecular assays, in vitro killing assays to interrogate CAR-T cell anti-tumor function, multi-color flow cytometry, and in vivo experimentation. In addition to designing and performing experiments, the candidate will also be expected to analyze, interpret, and present the data, as well as train new members of the lab.
